RESUMO

The emergence of Homo sapiens in Eastern Asia is a topic of significant research interest. However, well-preserved human fossils in secure, dateable contexts in this region are extremely rare, and often the subject of intense debate owing to stratigraphic and geochronological problems. Tongtianyan cave, in Liujiang District of Liuzhou City, southern China is one of the most important fossils finds of H. sapiens, though its age has been debated, with chronometric dates ranging from the late Middle Pleistocene to the early Late Pleistocene. Here we provide new age estimates and revised provenience information for the Liujiang human fossils, which represent one of the most complete fossil skeletons of H. sapiens in China. U-series dating on the human fossils and radiocarbon and optically stimulated luminescence dating on the fossil-bearing sediments provided ages ranging from ~33,000 to 23,000 years ago (ka). The revised age estimates correspond with the dates of other human fossils in northern China, at Tianyuan Cave (~40.8-38.1 ka) and Zhoukoudian Upper Cave (39.0-36.3 ka), indicating the geographically widespread presence of H. sapiens across Eastern Asia in the Late Pleistocene, which is significant for better understanding human dispersals and adaptations in the region.

The geographic expansion of Homo sapiens populations into southeastern Europe occurred by ∼47,000 years ago (∼47 ka), marked by Initial Upper Palaeolithic (IUP) technology. H. sapiens was present in western Siberia by ∼45 ka, and IUP industries indicate early entries by ∼50 ka in the Russian Altai and 46-45 ka in northern Mongolia. H. sapiens was in northeastern Asia by ∼40 ka, with a single IUP site in China dating to 43-41 ka. Here we describe an IUP assemblage from Shiyu in northern China, dating to ∼45 ka. Shiyu contains a stone tool assemblage produced by Levallois and Volumetric Blade Reduction methods, the long-distance transfer of obsidian from sources in China and the Russian Far East (800-1,000 km away), increased hunting skills denoted by the selective culling of adult equids and the recovery of tanged and hafted projectile points with evidence of impact fractures, and the presence of a worked bone tool and a shaped graphite disc. Shiyu exhibits a set of advanced cultural behaviours, and together with the recovery of a now-lost human cranial bone, the record supports an expansion of H. sapiens into eastern Asia by about 45 ka.

This transition from gathering to cultivation is a significant aspect of studying early agricultural practices. Fruit trees are an essential component of food resources and have played a vital role in both ancient and modern agricultural production systems. The jujube (Ziziphus jujuba Mill.), with its long history of cultivation in northern China, holds great importance in uncovering the diet of prehistoric humans and understanding the origins of Chinese agricultural civilization. This paper focuses on the domestication of jujube by analyzing the morphology of jujube stones found in three Neolithic sites in northern China's Qi River basin, Zhujia, Wangzhuang, and Dalaidian. The measurements of these jujube kernels are compared with those found in other areas of northern China, as well as modern jujube kernels that were collected. The measurements revealed that the length-to-diameter (L/D) ratio of sour jujube kernels ranged from 1.36 to 1.78, whereas the L/D ratio of cultivated jujube stones varied between 1.96 and 4.23. Furthermore, jujube stones obtained from Zhujia and Wangzhuang sites exhibit pointed ends and possess an elongated oval or narrow oval shape overall, which is indicative of clearly artificial domestication traits. Therefore, this study suggests that jujube was selected and cultivated as an important food supplement in the Qi River basin no later than around 6200 BP.

The automatic detection of cells in microscopy image sequences is a significant task in biomedical research. However, routine microscopy images with cells, which are taken during the process whereby constant division and differentiation occur, are notoriously difficult to detect due to changes in their appearance and number. Recently, convolutional neural network (CNN)-based methods have made significant progress in cell detection and tracking. However, these approaches require many manually annotated data for fully supervised training, which is time-consuming and often requires professional researchers. To alleviate such tiresome and labor-intensive costs, we propose a novel weakly supervised learning cell detection and tracking framework that trains the deep neural network using incomplete initial labels. Our approach uses incomplete cell markers obtained from fluorescent images for initial training on the Induced Pluripotent Stem (iPS) cell dataset, which is rarely studied for cell detection and tracking. During training, the incomplete initial labels were updated iteratively by combining detection and tracking results to obtain a model with better robustness. Our method was evaluated using two fields of the iPS cell dataset, along with the cell detection accuracy (DET) evaluation metric from the Cell Tracking Challenge (CTC) initiative, and it achieved 0.862 and 0.924 DET, respectively. The transferability of the developed model was tested using the public dataset FluoN2DH-GOWT1, which was taken from CTC; this contains two datasets with reference annotations. We randomly removed parts of the annotations in each labeled data to simulate the initial annotations on the public dataset. After training the model on the two datasets, with labels that comprise 10% cell markers, the DET improved from 0.130 to 0.903 and 0.116 to 0.877. When trained with labels that comprise 60% cell markers, the performance was better than the model trained using the supervised learning method. This outcome indicates that the model's performance improved as the quality of the labels used for training increased.

To investigate the effects of rumen-protected choline (RPC) and rumen-protected nicotinamide (RPM) on liver metabolic function based on transcriptome in periparturient dairy cows, 10 healthy Holstein dairy cows with similar parity were allocated to RPC and RPM groups (n = 5). The cows were fed experimental diets between 14 days before and 21 days after parturition. The RPC diet contained 60 g RPC per day, and the RPM diet contained 18.7 g RPM per day. Liver biopsies were taken 21 days after calving for the transcriptome analysis. A model of fat deposition hepatocytes was constructed using the LO2 cell line with the addition of NEFA (1.6 mmol/L), and the expression level of genes closely related to liver metabolism was validated and divided into a CHO group (75 µmol/L) and a NAM group (2 mmol/L). The results showed that the expression of a total of 11,023 genes was detected and clustered obviously between the RPC and RPM groups. These genes were assigned to 852 Gene Ontology terms, the majority of which were associated with biological process and molecular function. A total of 1123 differentially expressed genes (DEGs), 640 up-regulated and 483 down-regulated, were identified between the RPC and RPM groups. These DEGs were mainly correlated with fat metabolism, oxidative stress and some inflammatory pathways. In addition, compared with the NAM group, the gene expression level of FGF21, CYP26A1, SLC13A5, SLCO1B3, FBP2, MARS1 and CDH11 in the CHO group increased significantly (p < 0.05). We proposed that that RPC could play a prominent role in the liver metabolism of periparturient dairy cows by regulating metabolic processes such as fatty acid synthesis and metabolism and glucose metabolism; yet, RPM was more involved in biological processes such as the TCA cycle, ATP generation and inflammatory signaling.

Copper nanoparticles have attracted a wide attention because of their low cost and high specific surface area. At present, the synthesis of copper nanoparticles has the problems of complicated process and environmentally unfriendly materials like hydrazine hydrate and sodium hypophosphite that would pollute water, harm human health and may even cause cancer. In this paper, a simple and low-cost two-step synthesis method was used to prepare highly stable and well-dispersed spherical copper nanoparticles in solution with a particle size of about 34 nm. The prepared spherical copper nanoparticles were kept in solution for one month without precipitation. Using non-toxic l-ascorbic acid as the reducing and secondary coating agent, polyvinylpyrrolidone (PVP) as the primary coating agent, and NaOH as the pH modulator, the metastable intermediate CuCl was prepared. Due to the characteristics of the metastable state, copper nanoparticles were rapidly prepared. Moreover, to improve the dispersibility and antioxidant, the PVP and l-ascorbic acid were used to coat the surface of copper nanoparticles. Finally, the mechanism of the two-step synthesis of copper nanoparticles was discussed. This mechanism mainly relies on the two-step dehydrogenation of l-ascorbic acid to obtain copper nanoparticles.

Homo sapiens was present in northern Asia by around 40,000 years ago, having replaced archaic populations across Eurasia after episodes of earlier population expansions and interbreeding1-4. Cultural adaptations of the last Neanderthals, the Denisovans and the incoming populations of H. sapiens into Asia remain unknown1,5-7. Here we describe Xiamabei, a well-preserved, approximately 40,000-year-old archaeological site in northern China, which includes the earliest known ochre-processing feature in east Asia, a distinctive miniaturized lithic assemblage with bladelet-like tools bearing traces of hafting, and a bone tool. The cultural assembly of traits at Xiamabei is unique for Eastern Asia and does not correspond with those found at other archaeological site assemblages inhabited by archaic populations or those generally associated with the expansion of H. sapiens, such as the Initial Upper Palaeolithic8-10. The record of northern Asia supports a process of technological innovations and cultural diversification emerging in a period of hominin hybridization and admixture2,3,6,11.

Ancient DNA research has developed rapidly over the past few decades due to improvements in PCR and next-generation sequencing (NGS) technologies, but challenges still exist. One major challenge in relation to ancient DNA research is to recover genuine endogenous ancient DNA sequences from raw sequencing data. This is often difficult due to degradation of ancient DNA and high levels of contamination, especially homologous contamination that has extremely similar genetic background with that of the real ancient DNA. In this study, we collected whole-genome sequencing (WGS) data from 6 ancient samples to compare different mapping algorithms. To further explore more effective methods to separate endogenous DNA from homologous contaminations, we attempted to recover reads based on ancient DNA specific characteristics of deamination, depurination, and DNA fragmentation with different parameters. We propose a quick and improved pipeline for separating endogenous ancient DNA while simultaneously decreasing homologous contaminations to very low proportions. Our goal in this research was to develop useful recommendations for ancient DNA mapping and for separation of endogenous DNA to facilitate future studies of ancient DNA.

The oasis villages of the Tarim Basin served as hubs along the ancient Silk Road, and they played an important role in facilitating communication between the imperial centers of Asia. These villages were supported by an irrigated form of cereal farming that was specifically adapted to these early oasis settlements. In this manuscript, we present the results from new archaeobotanical analyses, radiocarbon dating, and organic carbon isotopic studies directly from carbonized seeds at the Wupaer site (1500-400 BC) in the Kashgar Oasis of the western Tarim Basin. Our results showed that early farming in the oasis relied on a mixed wheat and barley system, but after 1200 BC was intensified through more elaborate irrigation, the introduction of more water-demanding legumes, and possibly a greater reliance on free-threshing wheat. These crops and the knowledge of irrigated farming likely dispersed into the Tarim Basin through the mountains from southern Central Asia. Improved agricultural productivity in the Tarim Basin may also have led to demographic and socio-political shifts and fed into the increased exchange that is colloquially referred to as the Silk Road.

Wheat and barley evolved from large-seeded annual grasses in the arid, low latitudes of Asia; their spread into higher elevations and northern latitudes involved corresponding evolutionary adaptations in these plants, including traits for frost tolerance and shifts in photoperiod sensitivity. The adaptation of farming populations to these northern latitudes was also a complex and poorly understood process that included changes in cultivation practices and the varieties of crops grown. In this article, we push back the earliest dates for the spread of wheat and barley into northern regions of Asia as well as providing earlier cultural links between East and West Asia. The archaeobotanical, palynological and anthracological data we present come from the Tongtian Cave site in the Altai Mountains, with a punctuated occupation dating between 5,200 and 3,200 calibrated years BP, coinciding with global cooling of the middle-late Holocene transition. These early low-investment agropastoral populations in the north steppe area played a major role in the prehistoric trans-Eurasian exchange.

Based on chronological and archaeobotanical studies of 15 Neolithic and Bronze Age sites from the northern Chinese Loess Plateau and southern Inner Mongolia-the agro-pastoral zone of China-we document changes in the agricultural system over time. The results show that wheat and rice were not the major crops of the ancient agricultural systems in these areas, since their remains are rarely recovered, and that millet cultivation was dominant. Millet agriculture increased substantially from 3000 BC-2000 BC, and foxtail millet evidently comprised a high proportion of the cultivated crop plants during this period. In addition, as the human population increased from the Yangshao to the Longshan periods, the length and width of common millet seeds increased by 20-30%. This demonstrates the co-evolution of both plants and the human population in the region. Overall, our results reveal a complex agricultural-gardening system based on the cultivation of common millet, foxtail millet, soybeans and fruit trees, indicating a high food diversity and selectivity of the human population.
